Apparent digestibility coefficient of nutrients from shrimp, mussel, diatom and seaweed by juvenile Holothuria scabra Jaeger The ability of Holothuria scabra to digest nutrients, such as organic matter (OM), protein and carbohydrate from animal and plant feed ingredients was investigated. Four test feeds prepared by mixing sand with single ingredients from animal sources (shrimp and mussel) and plant sources (diatom and seaweed) were fed to H. scabra to estimate apparent digestibility coefficient (ADC). The total assimilated nutrient (TAN) increased with ADC, whereas ingestion rate (IR) varied slightly among the feeds suggesting that ADC might be a good indicator of nutrient availability to H. scabra. The ADCOM of shrimp and mussel was significantly higher than that diatom and seaweed: 86.2%, 77.1%, 55.1% and 32.3% respectively. ADCprotein was similar for shrimp (88.7%), mussel (84.8%) and diatom (75.2%), but significantly lower in seaweed (34.4%). ADCcarbohydrate was similar in mussel (58.5%) and diatom (58.3%) as well as in seaweed (31.6) and shrimp (28.0%). ADCprotein was relatively higher than ADCcarbohydrate suggesting that H. scabra generally digests more protein than carbohydrate. Furthermore, results indicated that nutrients from animal-based feeds are more efficiently digested by H. scabra; thus, animal ingredients rich in easily digestible protein could potentially provide an efficiently balanced diet for H. scabra fed with diatom containing high easily digestible carbohydrate.

Environmental deterioration such as hypoxia and sulfide production due to water and sediment eutrophication originating from aquaculture effluents have been problematic, resulting to sporadic disease outbreaks and fish kills in the Philippines. Integrated multi-trophic aquaculture (IMTA) is one of the promising methods for sustainable aquaculture as it also provides a supplementary source of income to the fish farmers. IMTA is a polyculture system that integrates culturing of fed species (e.g. finfish) the main commodity, organic extractive species (e.g. deposit and filter feeding benthos) and inorganic extractive species (e.g. seaweed). In this study, IMTA techniques were established for small-scale coastal fish farmers in the Philippines, with sea cucumber (Holothuria scabra, commonly known as sandfish), as the key species. Sandfish commands the highest price in tropical sea cucumber species. Nitrogen (N) budget of sandfish in polyculture with milkfish (Chanos chanos) and Elkhorn sea moss (Kappaphycus alvarezii), both of which are commonly cultured in the Philippines, was estimated using a simple closed box model. Information on stocking density, stocking size, mortality, growth, feed ration, feed assimilation, NH4-N production and NH4-N absorption of these species was obtained from a series of experiments and existing literature. Culture conditions were as follows: 26 g milkfish were cultured in a 5 x 5 x 4 m cage at an average stocking density of 36.7 ind/m3 (i.e. usual practice in the Philippines) with an initial feeding ration of 10% of body weight which was gradually decreased to 4% over time; 10 g sandfish were cultured in a 5 x 5 x 0.3 m cage hung under the milkfish cage to trap particulate N waste (i.e. feces and leftover feed) from milkfish culture at a stocking density of 35 ind/m2; the stocking weight of Elkhorn sea moss line culture was 10 kg. The culture period was 200 days. It was estimated that milkfish culture under the above-mentioned schemes cumulatively produced 145 kg of particulate N, and milkfish and sandfish together excreted 60 kg of NH4-N in 200 days of culture. Daily assimilation rate of the particulate N by sandfish ranged from 3.4 to 12.4%, and 6.4% of the particulate N was estimated to be removed by sandfish during the entire 200 days of culture. Daily absorption rate of NH4-N by Elkhorn sea moss increased exponentially with time and reached 100% at 125 days of culture. Cumulative NH4-N from milkfish and sandfish excretion was estimated to be depleted by 162 days of culture. For complete utilization of particulate N by sandfish by the end of milkfish culture period (i.e. zero emission), sandfish stocking density should be 805 ind/m2, which is 200 times as high as that in existing sandfish aquaculture operations in countries such as Viet Nam and New Caledonia. The purpose of sandfish culture in IMTA should be emphasized in terms of its economic advantages and not very much on environmental integrity. Cages for sandfish culture should be designed in such a way where only a small fraction of organic matter from milkfish culture (i.e. about 6% in this culture scheme) enters it to avoid sediment quality deterioration and possible death of sandfish. Elkhorn sea moss on the other hand seems very efficient in bioremediation capability.
Lipids and fatty acids in wild and pond-reared mud crab Scylla serrata (Forsskål) during ovarian maturation and spawning VR Alava, ET Quinitio, JB De Pedro, FMP Priolo, ZGA Orozco & M Wille -
Aquaculture Research, 2007 - Wiley-BlackwellWild-caught and pond-reared female mud crab Scylla serrata at different stages of ovarian maturation were collected from Samar and Capiz, Philippines. Crabs were categorized into five stages according to the external morphological and microscopic appearance of the most advanced oocytes. The ovaries, hepatopancreas, muscle and newly spawned eggs (NSE) were analysed for lipid class components and fatty acids. Total lipid was higher in pond-reared than in wild-caught crabs but increased with ovarian maturation in both groups. Ovarian lipid peaked at the fully mature stage, coinciding with a decline in hepatopancreatic and muscle lipids. Lipid levels declined significantly in spent females. The tissues contained elevated highly unsaturated fatty acids such as arachidonic (20:4n-6), eicosapentaenoic (20:5n-3) and docosahexaenoic (22:6n-3) acids, but at higher levels in late maturing and fully mature ovaries and in NSE. The type of lipid class and fatty acid components in mature ovaries as well as in NSE are generally considered to be indicative of their importance in reproductive physiology and embryonic and larval development.

The Israeli Journal of Aquaculture-Bamidgeh, 2009 - Society of Israeli Aquaculture and Marine BiotechnologyPreventing the transboundary movement of fish viral pathogens in a global environment requires active surveillance. This study examined the presence of three emerging viral pathogens among koi, common, grass, and silver carp in Cambodia, Lao PDR, Myanmar, Philippines, and Vietnam. The studied viruses included koi herpesvirus (KHV), spring viremia of carp virus (SVCV), and grass carp reovirus (GCRV). Detection methods consisted of virus isolation by cell culture, infection assay in naive fish, polymerase chain reaction (PCR), and reverse-transcriptase PCR (RT-PCR). Tissues were collected and pooled from 193 fish samples in Dec. 2004 to Feb. 2005, 406 in Sep. 2005 to Feb. 2006, and 1302 in Oct. 2006 to Feb. 2007. For cell culture, tissue filtrates were prepared from pooled spleens, kidneys, livers, and gills and inoculated onto koi fin (KF-1), grass carp kidney (GCK), and fat head minnow (FHM) cells. For infection assay, tissue filtrates were injected intraperitoneally to healthy, naive common carp. No virus was detected after three cell culture passages and the infection bioassays. One-step and nested-step PCR was used to detect KHV in gills of fish samples. One-step and semi-nested RT-PCR was used to detect SVCV and GCRV in the spleens, kidneys, and livers of fish samples. Samples from all three years from all five countries yielded negative results for all three viruses, indicating that KHV, SVCV, and GCRV were not present in these five countries during the period of the study although KHV outbreaks had been detected in Indonesia, Taiwan, Japan, Thailand, China, and Malaysia.
