Abstract
In this study, the Bacillus sp. JL47, a superior PHB-accumulating Bacillus strain identified from the previous work, was tested for its protective effects in gnotobiotic Artemia franciscana during a pathogenic Vibrio campbellii challenge. The utilization of gnotobiotic Artemia is important in this experiment because any possible microbial interference (which are naturally present in conventional culture system) are eliminated in this model culture system, hence the interpretation of the results in this mechanistic study can be more conclusive. The survival of the Artemia fed the Bacillus sp. JL47 at 1 × 107 cells mL–1 was significantly higher as compared with the challenged control and the survival was almost doubled when the dose was increased to 5 × 107 cells mL−1. However, feeding the Artemia at 106 cells mL−1 or lower showed no significant protective effects. Based on these densities, the estimated concentration of amorphous PHB showing a significant protection in Artemia was c. 2.44 mg L−1 and the effects were even better when the amorphous PHB level was increased to 12.19 mg L−1. Furthermore, feeding Bacillus sp. JL47 containing 55% amorphous PHB (on CDW) to Artemia showed a significantly higher survival in a Vibrio challenge relative to Bacillus containing 29% PHB. The data suggest that the amorphous PHB accumulated in the Bacillus sp. JL47 strain is an important determinant for the increased survival of challenged Artemia.

    Quantitative and qualitative analyses of the bacterial microbiota of tilapia (Oreochromis niloticus) cultured in earthen ponds in the Philippines

    The quantity and composition of the bacterial microbiota in the rearing water, sediment, gills and intestines of tilapia Oreochromis niloticus collected every 2 weeks from Day 30 to Day 120 after stocking for grow-out culture in 6 earthen brackish water ponds in the Philippines were examined. The total heterotrophic aerobic bacterial counts obtained in the water, sediment, gills and intestines of tilapia ranged from 103 to 104 c.f.u. ml−1, 103–105, 105–107 and 104–107 c.f.u. g−1, respectively. In terms of composition, a total of 20 bacterial genera and 31 species were identified with the preponderance of gram-negative bacteria constituting 84% of all bacterial isolates examined. Aeromonas hydrophila, Bacillus spp., Plesiomonas shigelloides, Shewanella putrefaciens, Pseudomonas fluorescens, Staphylococcus spp. and Vibrio cholerae were the dominant bacteria identified in the gills and intestine of tilapia. These bacteria also dominated in the pond sediment and rearing water, except for the nil isolation of S. putrefaciens and V. cholerae in the water samples examined, indicating that resident bacteria in the pond water and sediment congruently typify the composition of bacterial microbiota in the gills and intestine of tilapia which under stressful conditions may propel the ascendance of disease epizootics.

    In vitro and in vivo evaluation of the efficacies of commercial probiotics and disinfectant against acute hepatopancreatic necrosis disease and luminescent vibriosis in Litopenaeus vannamei

    The bioactivities of two commercially available probiotics and one chemical disinfectant were tested against strains of Vibrio parahaemolyticus (VPAHPND) and V. harveyi. This study aimed to determine shrimp pathogenic Vibrios' in vitro and in vivo sensitivities to commercial probiotics and a chemical disinfectant. The probiotics and disinfectant were tested first in vitro, followed by the in vivo trials. Results showed that upon administration of probiotics either through diet or adding into the tank water, the survivability of shrimp was increased during challenge with VPAHPND and V. harveyi. Also, the disinfectant was tested against the same pathogens and showed positive bactericidal effects at 2500 ppm and 5000 ppm. The present findings suggest that adding probiotics to the rearing water or the shrimp feeds effectively prevents infection by lowering the load of pathogenic bacteria. In comparison, the effectiveness of the disinfectant (PUR) depends on its appropriate concentration and timing of application. It is not only limited to rearing water but is also applicable for decontaminating pond liners, tanks, and other paraphernalia.

    Bacterial microbiota of hatchery-reared freshwater prawn Macrobrachium rosenbergii (De Man, 1879) and their pathogenicity

    Quantitative and qualitative analyses of bacterial microbiota associated with hatchery of Macrobrachium rosenbergii (de Man, 1879) were conducted over three larval cycles, along with important water quality parameters. Physicochemical parameters (temperature, DO, salinity, pH, and total ammonia) of culture water were within the optimum level required for the hatchery phase of M. rosenbergii. Significant differences in aerobic plate count (APC) among larval stages were observed. Ten genera and 17 species were identified with the predominance of Gram-negative bacteria constituting 63 % of all isolates (n = 706). A total of 17 dominant bacterial isolates randomly selected from freshwater prawn larvae were screened for in vitro hemolysin activity using blood agar plate (BAP) medium with 5% sheep blood. Pathogenicity of the isolated bacterial strains was assessed through in vivo infection bioassay. Out of the 17 isolates tested, six isolates showed α – β zone of hemolysis on the BAP, i.e., Aeromonas caviae, Bacillus cereus, Plesiomonas shigelloides, Staphylococcus warneri, Vibrio cholerae non-01, and Vibrio furnissii, were used in the infection bioassay. The current data indicate that the six bacterial isolates tested, particularly B. cereus and V. cholerae non-01, are opportunistic pathogens capable of inducing disease epizootics, particularly in freshwater prawn hatcheries with suboptimal rearing water conditions. Thus, control measures to reduce the influx of pathogenic microbes in the system by maintaining good water quality and good farm management practices through disinfection of culture facilities, rearing water, and Artemia cysts, the practice of good hygiene of personnel, regular water exchange, and feed regulation, among others, and cautionary use of antibiotics, can be adopted.
