Growth promotion and enhancement of vibriosis resistance in penaeid shrimp after oral administration of lactic acid bacteria and marine bacteria

Vibriosis is one of the most serious problems in the shrimp culture industry worldwide. Antibiotic treatment is becoming less and less effective, as new strains of pathogens evolve, which are resistant to commonly used antibiotics. Use of probiotics is an alternative method for controlling bacteria in shrimp culture. In this experiment, lactic acid bacteria (Peddiococcus sp, Lactobacillus sp, and L. acidophilus) and marine bacteria (Alteromonas sp. and Vibrio alginolyticus) were mixed with pellet feed and used to feed juvenile shrimp. The growth and survival rate of the shrimp were monitored during the experiment. After feeding the shrimp for 60 days, all the surviving shrimp were bath-challenged with Vibrio harveyi (105-106 CFU/ml) for 7 days. Mortality of the shrimp was recorded. The result shows that the total biomass and survival rate of the shrimp fed with probiotics were significantly higher than the control group (p<0.05). However, there were no significant differences in the mean growth rate of shrimp between treatments. The results of the challenge test shows that the groups fed with lactic acid bacteria had the highest survival rates (Peddiococcus sp., 86.4%; Lactobacillus sp., 90.5% and L. acidophilus, 93.8%), followed by the treatments fed with marine bacteria (Alteromonas sp, 74.6% and Vibrio alginolyticus, 61.8%) and the control treatment had the lowest survival rate (59%).

    Bacterial microbiota of hatchery-reared freshwater prawn Macrobrachium rosenbergii (De Man, 1879) and their pathogenicity

    Quantitative and qualitative analyses of bacterial microbiota associated with hatchery of Macrobrachium rosenbergii (de Man, 1879) were conducted over three larval cycles, along with important water quality parameters. Physicochemical parameters (temperature, DO, salinity, pH, and total ammonia) of culture water were within the optimum level required for the hatchery phase of M. rosenbergii. Significant differences in aerobic plate count (APC) among larval stages were observed. Ten genera and 17 species were identified with the predominance of Gram-negative bacteria constituting 63 % of all isolates (n = 706). A total of 17 dominant bacterial isolates randomly selected from freshwater prawn larvae were screened for in vitro hemolysin activity using blood agar plate (BAP) medium with 5% sheep blood. Pathogenicity of the isolated bacterial strains was assessed through in vivo infection bioassay. Out of the 17 isolates tested, six isolates showed α – β zone of hemolysis on the BAP, i.e., Aeromonas caviae, Bacillus cereus, Plesiomonas shigelloides, Staphylococcus warneri, Vibrio cholerae non-01, and Vibrio furnissii, were used in the infection bioassay. The current data indicate that the six bacterial isolates tested, particularly B. cereus and V. cholerae non-01, are opportunistic pathogens capable of inducing disease epizootics, particularly in freshwater prawn hatcheries with suboptimal rearing water conditions. Thus, control measures to reduce the influx of pathogenic microbes in the system by maintaining good water quality and good farm management practices through disinfection of culture facilities, rearing water, and Artemia cysts, the practice of good hygiene of personnel, regular water exchange, and feed regulation, among others, and cautionary use of antibiotics, can be adopted.

    Efficiency of Chlorella sp. and Tilapia hornorum in controlling the growth of luminous bacteria in a simulated shrimp culture environment

    This study clarifies the effect of green water from Chlorella sp., Tilapia hornorum and the combination of the two organisms on the growth of luminous bacteria in a simulated shrimp culture environment. Results showed that the presence of Chlorella sp. (105 cells/ml) alone was not effective in the control of luminous bacteria in shrimp (biomass=80 g/m3) rearing water. The presence of T. hornorum alone (biomass=500 g/m3) was more efficient in controlling the growth of luminous bacteria than the co-existence of tilapia and Chlorella sp. Nevertheless, the presumptive Vibrio count was lowest in control tanks that had the highest shrimp survival rate, which was attributed to the presence of other micro-algae such as Chaetoceros, Thalassiosira, Navicula, Nitszchia, Melosira, and Fragilaria.

    Level and percentage recovery of resistance to oxytetracycline and oxolinic acid of bacteria from shrimp ponds

    The bacterial level of the water, sediment and cultured shrimp (Penaeus monodon) from different ponds were determined using a general medium, a presumptive Vibrio medium and a presumptive Pseudomonas–Aeromonas medium. Samples were taken from ponds that had not used any antimicrobial, ponds that had previously used and also ponds that were currently using oxolinic acid (OXA). The bacterial level in the sediment was higher than in the water using all three media. More bacteria existed in the pond system than in the receiving environment. Shrimp hepatopancreas harbored more bacteria than the lymphoid organ. The Vibrio density of the pond and bacterial levels in the shrimp were correlated with the use of the antimicrobial. The Vibrio level of the pond samples and microbial density of shrimps were higher from ponds that had not used any antimicrobials. The percentage recoveries of resistance to oxytetracycline (OTC) and OXA in bacteria from shrimp ponds and cultured shrimps were also determined using Zobell's marine agar, Pseudomonas–Aeromonas selective agar and thiosulfate citrate bile sucrose agar (TCBS) with the addition of either 25 μg/ml OTC or 25 μg/ml OXA. Presumptive Vibrio bacteria and other bacterial taxa recovered from the pond/receiving water/sediment from all three sites showed some degree of resistance to OTC and OXA. However, a higher percentage recovery of strains resistant to OTC than to OXA was observed among the presumptive vibrios and other bacterial taxa. Pseudomonas and Aeromonas bacteria were more resistant to OXA compared with the vibrios. All bacterial taxa resistant to OTC were more readily recovered from the water samples than from the sediment samples. In general, between the samples from the pond and from the receiving environment, a higher percentage of resistant strains was observed in the latter. Higher percentage recovery of bacteria resistant to OXA was observed in shrimp from ponds currently using OXA than those from ponds that had not used any or those that had previously used them. The results of the present study showed that the percentage recovery of resistance reflected the pattern of antimicrobial use.
