Effects of intraperitoneal injections of LHRH-a and domperidone (DOM), given singly or in combination at two injections, on oocyte maturation, ovulation and spawning in bighead carp, Aristichthys nobilis, were investigated. Results were compared with the existing protocol using HCG + LHRH-a at 1800 IU and 20 μg/kg BW, respectively, to induce spawning in bighead carp. Oocyte diameter of fish at 6 h after the second injection of LHRH-a + DOM and HCG + LHRH-a had significantly increased to 7.0% and 7.5% respectively, as compared to the 0 h group (P<0.05); fish injected with LHRH-a, DOM or saline had significantly lower increases (1.0%, 2.0%, and 3.0%, respectively). Ovulation was induced in 75% and 60% of fish injected with LHRH-a + DOM and HCG + LHRH-a, respectively. In contrast, fish treated with LHRH-a, DOM or saline did not ovulate. Total eggs spawned, and fertilization and hatching rates did not differ significantly (P<0.05) between the two groups with ovulated fish. Although injection protocols using LHRH-a + DOM and HCG + LHRH-a were equally effective in inducing spawning of bighead carp, the former had a lower combined cost than the latter.

LHRHa and pimozide-induced breeding in the catfish, Clarias macrocephalus (Gunther)

Induced breeding protocol was developed by monitoring the size and maturation of eggs at 0-48 h after a simultaneous injection of luteinizing hormone-releasing hormone analogue (LHRHa; 0.0005 - 0.10 µg/g BW) and pimozide (PIM; 1 µg/g BW). Based on its similar osmotic pressure with catfish plasma, eggs were fixed in 1% phosphate-buffered formalin. Mean egg diameter of fish that were induced to mature increased during ovulation. Oocyte maturation, indicated by oocytes with germinal vesicle breakdown (GVBD), was observed at least 12 h post-injection in fish given 0.01 - 0.10 µg LHRHa + 1 µg PIM/g BW, followed by ovulation 4 h thereafter. Results showed that a simultaneous injection of C. macrocephalus with 0.05 µg LHRHa + 1 µg PIM/g BW at 1800-1900 h followed by stripping at 16-20 h post-injection resulted in high ovulation, fertilization and hatching rates. -
Effects of single injection of carp pituitary extract and human chorionic gonadotropin on germinal vesicle migration and ovulation in lampan sungai, Puntius schwanenfeldii (Bleeker): A preliminary study

(Aquaculture Department, Southeast Asian Fisheries Development Center, 1996)The effects of crude pituitary extract (CPE) and human chorionic gonadotropin (hCG) on germinal vesicle migration and ovulation in lampan sungai Puntius schwanenfeldii (Bleeker) were investigated. Sexually mature female lampan sungai were divided into four groups (n=3 per group). In both experiments, fish were injected intramuscularly (im) once and were checked for germinal vesicle migration and sign of ovulation 8 h following injection. In experiment 1, females were treated with CPE at doses of 2, 4, or 6 mg/kg body weight (BW) while controls were administered an equivalent volume of saline. Ovulation occurred in one fish given 2 mg/kg and in another fish that received 6 mg/kg CPE. In a second experiment, female fish were administered hCG at doses of 1000, 2000, or 4000 IU/kg body weight. Similarly, controls received only saline. One female injected with 4000 IU/kg hCG responded positively to the hormone treatment. Clearly, in our study, a single intramuscular injection of CPE and hCG induced spawning in lampan sungai as well as accelerated oocyte development as evidenced by the increasing percentage of oocytes showing germinal vesicle migration. -
Effect of season on oocyte development and serum steroid hormones in LHRHa and pimozide-injected catfish Clarias macrocephalus (Günther)
Tan-Fermin, Josefa D.; Marte, Clarissa L.; Ueda, Hiroshi; Adachi, Shinji; Yamauchi, Kohei (Japanese Society of Fisheries Science, 1999)Oocyte and blood samples were taken from gravid female catfish Clarias macrocephalus at 4-h intervals to monitor the stage of oocyte development and serum steroid hormone profiles after injection of luteinizing hormone-releasing hormone analogue (LHRHa) and pimozide (PIM) during the off-season (February) and the peak of the natural breeding period (August). Results showed that the onset of final oocyte maturation (12h) and ovulation (16h), and levels of serum estradiol-17β (E2) did not vary with season in LHRHa+PIM-injected fish. In February, ovulated eggs were stripped from three and two hormone-treated fish at 16h and 20h post-injection, respectively. In August, ovulation was observed in all hormone-treated females (n=5) at 16h post-injection but stripping of the eggs was possible only 4h thereafter. Serum E2 levels were significantly different only with varying time post-injection; a marked increase occurred at 12h, but the elevation was higher in fish induced to ovulate during the peak (16.8ng/ml) than off-season (7.7ng/ml). Hormone-treated fish showed higher serum testosterone (T) levels during the peak season (17-23ng/ml) than those injected during the off-season (10-20ng/ml) at 4-12h post-injection. Serum 17α, 20β-dihydroxy-4-pregnene-3-one (DHP) levels of hormone-treated fish during the off-season were only about half the level (0.29 and 0.52 ng/ml) of those treated with the same hormones during the peak season (0.54 and 0.9ng/ml) at 8 and 12h postinjection, respectively. Development of oocytes and serum steroid hormone profiles after LHRHa+PIM-induced ovulation provide basic understanding of the processes that mediate final oocyte maturation and ovulation in captive C. macrocephalus.
