Abstract
To estimate the portal of entry of Vibrio penaeicida in kuruma prawn (Penaeus japonicus), immersion and oral infection experiments were conducted with pre-treated and intact prawns. In immersion challenge, V. penaeicida produced 30% mortality in the shell-removed group at both 108 and 107 CFU/ml doses while in the eye-cut group, mortalities were 44% and 13% at 108 and 107 CFU/ml, respectively. In the intact group, 10% mortality was observed at 108 CFU/ml and no mortality at 107 CFU/ml after two weeks. The pathogen produced high mortalities in the orally incubated group, the median lethal dose (LD50) being 103-104 CFU/animal. In the groups given V. penaeicida-inoculated kuruma prawn meat and V. penaeicida-contaminated commercial pellets, mortalities within 60 days were 10% and 20%, respectively. These results suggest that the main portal of entry of V. penaeicida is the gastro-intestinal tract although the cuticle (epidermis) or wounds can also be portals.

    Bacterial flora in the hepatopancreas of pond-reared Penaeus monodon juveniles with luminous vibriosis

    Quantification and characterization of bacterial flora in the hepatopancreas (hp) of pond-reared Penaeus monodon juveniles affected with luminous bacteria were conducted in 1994 and 1995. Shrimp samples were taken from 23 grow-out ponds, 14 of which had disease outbreaks. Luminous bacterial (LB) load of the shrimps' hp with (mean=2.4×101 colony forming units (CFU)/hp) and without (mean=0.3×101 CFU/hp) disease outbreaks were comparable during the first 15 days of culture (DOC). During disease outbreaks at 18 to 32 DOC, however, LB load of affected shrimps (mean=9.0×104 CFU/hp) were higher than healthy shrimps (mean=7.0×101 CFU/hp). At 50 to 60 DOC, levels of LB were comparable in older shrimps with or without disease. Total viable and presumptive Vibrio counts were also comparable in both shrimp samples from 1 to 60 DOC. Characterization of the 172 bacterial isolates collected showed that most (90.12%) were Vibrio species dominated by V. harveyi (27.91%), V. splendidus II (13.37%) and V. parahaemolyticus (10.46%).

    Research on crustaceans

    Crustacean research at the SEAFDEC Aquaculture Department during the last three years focused mostly on the tiger shrimp Penaeus monodon. Studies were done along six problem areas: (1) developing spawning techniques for captive broodstock, (2) defining physico-chemical levels tolerable by larvae or postlarvae, (3) finding alternative feeds or fertilizers for extensive culture, (4) reducing the cost and evaluating the quality of formulated feeds for semiintensive culture, (5) preventing and controlling disease, and (6) documenting the chemicals used in shrimp culture and their effects on the environment. To reduce feed costs, substitutes for expensive feed components were screened and the specific nutrient requirements of tiger shrimp during culture were determined. A few studies were made on other crustaceans. The vitellogenin levels during maturation of the white shrimp P. indicus were measured. The digestibility of feedstuffs was also tested in the white shrimp. Culture techniques are being developed for the mudcrab Scylla serrata in ponds, pens, and cages.

    Effect of Epinephelus coioides, Chanos chanos, and GIFT tilapia in polyculture with Penaeus monodon on the growth of the luminous bacteria Vibrio harveyi

    Studies have shown that the presence of Tilapia hornorum hybrid has antibacterial effect against luminous bacteria. The present study aims to determine the effect of different fish species such as grouper, milkfish and tilapia in polyculture with shrimp on the growth of luminous bacteria. Results showed that stocking of tilapia Oreochromis niloticus hybrid and grouper Epinephelus coioides at a biomass of 500 g/m3 efficiently inhibited the growth of luminous bacteria in shrimp (biomass = 80 g/m3) rearing water and positively affected shrimp survival. Results also showed that the presence of milkfish Chanos chanos at a biomass of 500 g/m3 did not inhibit the growth of luminous bacteria in shrimp (biomass = 80 g/m3) rearing water.
