Genetic research initiatives for sustainable aquaculture production in the Philippines
(Field Science Center, Graduate School of Agricultural Science, Tohoku University, 2019-03)The Philippines is one of several countries in Southeast Asia that has, for several decades, made steady contributions to world aquaculture production both from inland and marine waters. In recent years, fish production has been on the decline mainly because of the lack of quality seedstock, limited stocks of captive breeders or spawners of major aquaculture commodities, adverse effects of climate change and other environmental factors on fish breeding and rearing, fish diseases caused by pathogenic organisms and prohibitive cost of aquaculture inputs such as feeds, etc. Genetic researches have been conducted mostly through local grants with the aim of addressing the aforementioned constraints. Such initiatives focused on developing and applying methods in (a) selective breeding; (b) marker-assisted genetic strain assessment for broodstock development and for monitoring inbreeding in farmed stocks and (c) genomics to understand and enhance on-farm stock performance through the identification of genes that are responsible for nutrition, stress and immune responses, among others. This paper highlights examples of local genetics applications in tilapia, mangrove crab, shrimp, milkfish and abalone aquaculture. The significance of implementing genetic interventions to boost and sustain aquaculture production in the Philippines is likewise discussed. -
Preliminary assessment of genetic variation in the Japanese endemic freshwater crab, Geothelphusa dehaani, based on mitochondrial DNA sequences
(Pensoft Publishers, 2023-06-27)Geothelphusa dehaani, a freshwater crab species endemic to Japan, has the largest distribution range amongst the 19 known species in the country. Due to its low dispersal capability and restricted habitat to freshwater, it serves as an excellent model for understanding gene flow between geographically isolated populations. In this study, we analysed the genetic relationships of 26 G. dehaani populations collected from different locations in the Japanese archipelago using two mitochondrial DNA regions - cytochrome oxidase subunit I (COI) and cytochrome b (cytB). Our results from the analysis of molecular variance (AMOVA) revealed high genetic variation amongst populations and the phylogenetic analysis identified four geographical groups: Clade I - Honshu and Shikoku, Clade II - north-eastern Kyushu, Clade III - southern Kyushu and Clade IV - north-western Kyushu. Notably, Clade IV exhibited the highest genetic distance amongst the observed groupings. These findings highlight the need for further examination of G. dehaani in Kyushu, including morphological and behavioural traits, to better understand the observed diversity within the species in the region. -
Reproductive quality evaluation of male Indian white prawn Penaeus indicus broodstock-fed diets supplemented with polychaete extracts (Marphysa sp.)
(Taylor and Francis, 2023-06-23)The present study determined the effect of different polychaete extracts, namely, total soluble fraction (TSF), neutral lipid fraction (NLF) and polar lipid fraction (PLF), in the maturation and sperm quality of male Penaeus indicus. Three levels (0.25, 0.50 and 1.00%) of extracts were included using a 3 × 3 factorial design. Groups fed the basal diet (BD) and fresh-frozen diet served as controls. Extracts in varying doses and control groups did not have a significant effect on broodstock survival (67–87%; p = 0.960), maturation rate (42–68%; p = 0.615), inter-spermatophore period (8–10 days; p = 0.505) or sperm viability (97–100%; p = 0.819). However, sperm counts of broodstock fed BD (11.70 × 106 ±1.05 × 106 per spermatophore) and those fed diets supplemented with polychaete extracts were significantly higher compared to that fed with control fresh diet at 0.73 × 106± .09 × 106 (p = 0.001). Spermatophore crude lipid was highest in groups fed 0.25% TSF and 0.25% PLF of broodstock (p =1.0 x10−6 ). Inclusion of TSF (0.25–1.00%) significantly increased the spermatophore crude protein content of broodstock compared to those fed with other diets (p = 1.20 × 10−5). These results demonstrate that NLF and TSF extracts are bioactive components of polychaete which when fed to male P. indicus, can stimulate aspects of sperm production. -
Reproduction, early development, and larval rearing of the climbing perch Anabas testudineus (Teleostei: Anabantidae) in captivity
(College of Agriculture and Food Science, University of the Philippines Los Baños, 2023-06)Induced spawning, early development, and larval rearing of the climbing perch Anabas testudineus were examined under captive conditions. Five female climbing perch [125.33 ± 3.27 mm standard length (SL); 70.67 ± 5.59 g body weight (BW)] were each paired with apparently mature males (108.50 ± 3.97 mm SL; 39.27 ± 4.70 g BW) and induced to spawn with 5,000 IU human chorionic gonadotropin (hCG) + 0.5 mL Ovaprim/kg BW. All hCG + Ovaprim-injected fish spawned 9 – 11 h after hormone administration but saline-injected fish (control group) failed to spawn. Mean egg production per female, fertilization and hatching rates, and larval production per female were 7,667 ± 1,313, 98.11 ± 1.63%, 61.01 ± 19.92%, and 4,435 ± 1,041, respectively. Fertilized eggs (1.52 – 1.96 mm in diameter) were almost spherical, non-adhesive, transparent, and buoyant due to a large, single oil globule (515 ± 33 µm in diameter). First cleavage appeared 30 min post-fertilization (MPF) and egg development lasted for 20 – 24 h post-fertilization (HPF) (29°C). Newly hatched climbing perch larvae [1.73 ± 0.08 mm in total length (TL)] with yolk volume (YV) and oil globule volume (OGV) of 0.545 ± 0.104 mm3 and 0.072 ± 0.013 mm3, respectively had no pigmented eyes or functional mouth parts and digestive tract, and immobile with the yolk sac pointed upward. Eyes became pigmented and both mouth and anus opened in 1 d old larvae (2.09 ± 0.04 mm TL) in preparation for exogenous feeding. Except for body depth (BD) and pre-anal length (PAL), a general increase in TL (1.73 – 3.72 mm), eye diameter (ED) (0.164 – 0.378 mm), head length (HL) (0.205 – 0.923 mm), and mouth gape (MG) size (0.308 – 0.552 mm) was observed, coinciding with yolk resorption in 7 d post-hatched larvae. Climbing perch larvae were reared at four stocking densities (25, 50, 75, 100 larvae L-1) and fed solely on live food (Brachionus rotundiformis from days 2 to 12 and Artemia nauplii from days 13 to 40) or co-fed live food and microparticulate diet (MPD) (Artemia nauplii from days 2 to 15 and Artemia + MPD from days 16 to 40). Mean survival rates at lower densities (25 – 50 larvae L-1) were significantly higher than those reared at 75 and 100 larvae L-1 (P < 0.05). Furthermore, climbing perch larvae co-fed Artemia nauplii and MPD showed better survival (28.67 – 81.00%) than those fed solely on live food (24.88 – 64.00%). However, at the end of the 40 d trial period, growth parameters did not vary significantly among densities (P > 0.05), except for the condition factor in larvae co-fed Artemia nauplii and MPD. Taken together, results demonstrate the effectiveness of hCG + Ovaprim for captive breeding and observations on the early development of climbing perch, and the feasibility of rearing the larvae at 25 – 50 L-1 on a combination of Artemia nauplii and MPD under laboratory conditions. -
Behavior changes and \(\mathsf{LC_{50}}\) of dried garlic (Allium sativum) acute toxicity in Nile tilapia (Oreochromis niloticus) juvenile
(College of Agriculture and Food Science, University of the Philippines Los Baños, 2023-06)Plant-based compounds are emerging substitutes for chemical treatments in aquaculture since they are known to cause less harm to both the environment and its organisms. Garlic (Allium sativum) is a popular herb that has a broad range of uses, especially in the control and treatment of bacterial and parasitic diseases. However, its toxicity to tilapia is not well-studied. Hence, this study was conducted to test the toxicity levels of garlic powder in juvenile Nile tilapia (Oreochromis niloticus) by determining the cumulative mortality and median lethal concentration (LC50). Test fish were exposed through immersion in 6 concentrations (150, 200, 250, 300, 350, and 400 mg L-1) of garlic powder with 25% allicin and a control in 96 h static bioassay. Within 24 h, test fish exposed to 250 mg L-1 garlic and above, exhibited abnormal behaviors such as lethargy, gasping for air, and weak responses to environmental stimuli. Cumulative mortality was highest at 100% in 400 mg L-1 garlic at 48 h. Mortalities were dependent on the concentration and duration of exposure. The 96 h LC50 of garlic powder to O. niloticus is 225.86 mg L-1 with lower and upper confidence limits of 210.37 and 242.50 mg L-1, respectively at 27°C. -
Lime application to condition soil to pH 11 controls growth of harmful vibrios and WSSV
(European Association of Fish Pathologists, 2023-05-26)Shrimp farming has been devastated by several diseases. Disease outbreaks can be prevented by optimising environmental conditions or by eliminating risks factors including carrier organisms. These can be achieved through proper pond preparation. One important step in pond preparation is the application of hydrated lime. This paper describes the effect of hydrated lime application to condition pond soil to pH 11 on the pond ecosystem, more specifically on soil pH and biota. The experiment was done using three 600 m2 brackish water ponds. After hydrated lime application, the wet soil pH on the 1st-8th day was significantly higher (P<0.05) compared to the pH before liming to pH 11, and flushing after flooding for 24 h. Soil pH of 11 was observed 24-48h after lime application. Moreover, unwanted species like crabs and fish were found dead. Liming to pH 11 negatively affects the counts of green vibrio colonies thereby decreasing the percentage of green vibrios (PGV). In another experiment, three 750m2 earthen ponds were used. The pond soils were previously positive for the Whitespot Syndrome Virus (WSSV), hydrated lime were applied to bring the pond soil pH to 11. After lime application, WSSV was not detected in the pond soils. Results showed that increasing the soil pH to ≥11 positively affects the pond ecosystem. Liming encourages the proliferation of yellow vibrios and the eradication of carrier species (i.e. crabs) as well as predators (i.e. fish) in the ponds. Furthermore, liming to pH 11 may also eradicate White spot virus from the pond sediment. To implement a 1-unit increase in wet pH, hydrated lime should be applied at 2T/ha. -
Estimating the economic benefits and costs of conserving marine protected area in San Jose, Antique, Philippines
(Silliman University, 2023-03-27)The establishment of marine protected areas (MPAs) in the Philippines becomes a priority management strategy to conserve marine biodiversity and address the problem of declining marine resources. Economic studies can be a viable tool to determine MPA as a management option by evaluating their benefits and costs to the community. In order to determine the economic benefits of MPAs in San Jose, Antique, the total economic value of the resource consisting of direct and indirect use and non-use values was determined in 2016. Direct benefits from revenues and tourism amounted to Php 247,739.61 at the time of the study. Concurrently, the indirect functional benefits of MPA amounted to Php 2,162,235 using the benefit-transfer method. In measuring the non-use values of the MPA, the contingent valuation method was used to elicit the people’s willingness to pay for the MPA conservation program. The mean willingness to pay (WTP) of 260 participants equals Php 219.00, and the social WTP amounted to Php 5,725,136.89. The total benefits of managing the MPA amounted to Php 8,135,111.50, which exceeded the cost of conserving the MPA, which amounted to only Php 1,235,363.30. The net present values across 25 year period and different interest rates generated positive results. -
Histidine requirement of snubnose silver pompano Trachinotus blochii
(Elsevier, 2023-05-25)Histidine is necessary for the formation and repair of tissues to mitigate the effects of cataract on some fish species. Although the requirements for dietary amino acids of several Trachinotus species have been reported, the quantitative histidine requirement for a cost-effective diet for snubnose pompano or Trachinotus blochii, a high value finfish species in the Philippines, has been limited. A 90-day growth experiment determined the dietary histidine requirement of juvenile snubnose pompano (0.19±0.03 g). Six isonitrogenous, isolipidic, and isoenergetic diets consisted of a control diet with 0% histidine inclusion level (basal histidine level of 0.65%) and five diets with histidine inclusion levels of 0.15 (0.80%), 0.3 (1.1%), 0.45 (1.55%), 0.6 (2.15%), and 0.75% (2.9%). To simulate the pompano whole-body amino acid composition, essential amino acids were added in the diet. Fifteen pompano juveniles per tank in three replicates were stocked randomly in 250-L fiberglass tanks and were fed to satiation for 90 d. When the dietary histidine level and percent weight gain were analyzed via quadratic regression, a dietary histidine level of 1.61 g per 100 g diet or 0.357 g per 100 g protein is required for juvenile snubnose pompano. Moreover, the occurrence of cataracts was not observed in fish fed all dietary treatments. -
Molluscan aquaculture in the Philippines: A review
(Springer, 2023-04-25)Molluscs are among the most valuable resources of the Philippines, an archipelagic country of 7,107 islands bounded by 36,000 km of coastline and 26.6 million ha of coastal waters. In 2020, production from mussels and oysters alone valued at PhP1.552 billion. As primary sources of food and livelihood for marginalized fisherfolk, they are considered important in food security, nutrition, and poverty alleviation. Nevertheless, for almost a century, mollusc culture remained small-scale, with most fish farmers investing in non-fed species, like mussels and oysters. Moreover, mollusc culture in the country is still dependent on wild sources of seed stocks, making production unsustainable and unreliable. This dependence on the wild seed stocks will continue until the issue of the lack of commercial-scale hatcheries or the limited capacity of existing hatcheries to produce seeds, both for top and emerging mollusc aquaculture species, is resolved. Although technologies from broodstock management to post-harvest are available in some species, the lack of capital and the limited government support limit these artisanal fishers from doing large commercial-scale culture. This paper summarizes what has been done and published on the leading mollusc species cultured in the Philippines. Future research activities may be designed based on the research gaps to refine available technologies and develop new ones to improve culture production. The lack of or limited relevant information on the viable economics of the different culture techniques at different culture phases, the limited availability of post-harvest technologies, and the sparse or lack of genetic information for these leading mollusc species are among the gaps that need to be addressed. -
Grow-out culture of oyster Magallana bilineata (Röding, 1798) using pouches: A comparison of growth and survival in the river and earthen pond
(College of Agriculture and Food Science, University of the Philippines Los Baños, 2023-03)Oysters are among the most in-demand aquaculture commodities in the Philippines and worldwide. With the decreasing culture area and the deteriorating water quality of oyster beds, there is a need to explore new culture sites and techniques to address the problems of dwindling stocks and the increasing demand for better quality oysters. This study compared the growth and survival of the oysters Magallana bilineata, the new accepted name of Crassostrea iredalei, cultured in an earthen pond and the river using pouches suspended from rafts. Mean growth rates of oysters in length and weight were significantly higher in those reared in the river (0.56 ± 0.02 cm mo-1; 10.27 ± 0.42 g mo-1) than those in the pond (0.41 ± 0.03 cm mo-1; 5.99 ± 0.22 g mo-1), but the meat yield of oysters reared in the river (25.96 ± 0.92%) was almost the same as in the pond (24.05 ± 1.41%). Likewise, the proximate composition was the same for oysters coming from the river and the pond, respectively — 58.04 ± 0.14% and 53.86 ± 0.38% crude protein, 7.00 ± 0.08% and 7.93 ± 0.38% crude fat, 24.68 ± 0.07% and 28.54 ± 0.74% nitrogen-free extract, and 10.27 ± 0.01% and 9.68 ± 0.01% ash. Oyster survival at harvest was significantly higher in the river (70.21 ± 2.84%) than in the pond (13.10 ± 1.57%). However, with some interventions, ponds may still be utilized for oyster culture despite the low survival results in this study. Extending oyster culture in these aquaculture facilities may serve as a basis for interventions to make the pond more habitable for oysters and may help boost production in the country. -
Genomic analysis of Vibrio harveyi strain PH1009, a potential multi-drug resistant pathogen due to acquisition of toxin genes
(Elsevier, 2023-03-25)In has increasingly been observed that viral and bacterial coinfection frequently occurs among cultured shrimp and this coinfection could exacerbate the disease phenotype. Here, we describe a newly discovered bacterial strain, Vibrio harveyi PH1009 collected from Masbate Island, Philippines that was found to be co-infecting with the White Spot Syndrome virus a sample of black tiger prawn, Penaeus monodon. The genome of V. harveyi PH1009 was sequenced, assembled, and annotated. Average Nucleotide identity calculation with Vibrio harveyi strains confirmed its taxonomic identity. It is a potential multi-drug and multi-heavy metal resistant strain based on the multiple antibiotic and heavy metal resistance determinants annotated on its genome. Two prophage regions were identified in its genome. One contained genes for Zona occludens toxin (Zot) and Accessory cholera toxin (Ace), essential toxins of toxigenic V. cholerae strains apart from CTX toxins. Pan-genome analysis of V. harveyi strains, including PH1009, revealed an “open” pan-genome for V. harveyi and a core genome mainly composed of genes necessary for growth and metabolism. Phylogenetic tree based on the core genome alignment revealed that PH1009 was closest to strains QT520, CAIM 1754, and 823tez1. Published virulence factors present on the strain QT520 suggest similar pathogenicity with PH1009. However, PH1009 Zot was not found on related strains but was present in strains HENC-01 and CAIM 148. Most unique genes found in the PH1009 strain were identified as hypothetical proteins. Further annotation showed that several of these hypothetical proteins were phage transposases, integrases, and transcription regulators, implying the role of bacteriophages in the distinct genomic features of the PH1009 genome. The PH1009 genome will serve as a valuable genomic resource for comparative genomic studies and in understanding the disease mechanism of the Vibrio harveyi species. -
Effect of size grading on growth of yellow Pacific shortfin eel (Anguilla bicolor pacifica)
(Wiley, 2023-03-22)Heterogeneous growth, which is common among farmed fish, can be remedied by size grading. This study focused on whether the size grading process, which is commonly practiced in aquaculture, improves the subsequent growth performance of the Pacific shortfin eel Anguilla bicolor pacifica. Eels [338.70 ± 4.70 mm total length (TL) and 84.93 ± 1.87 g body weight (BW)] were initially reared for 60 days (pre-size grading period) and manually divided into three size groups: small (≤115 g), large (>150 g) and mixed (>115 g but <170 g), all maintained at a density of 5 kg m−3 and reared until day 150 (size grading period). After size grading, mean BW and TL at harvest significantly differed in all eel groups, suggesting growth was size specific. Nonetheless, at day 150, except for biomass, size grading had no significant effect on growth and survival, coefficient of variation of length and weight, and Fulton's condition factor. Large eels had a significantly higher feed intake than mixed or small eels, whereas feed efficiency did not differ among eel groups. Results suggest that size grading promotes size-specific growth but did not improve the overall growth performance of yellow stage of A. bicolor pacifica. -
Inclusion of live mud polychaete (Marphysa iloiloensis) in the feeding regime improved the hatchery performance of domesticated Indian white shrimp (Penaeus indicus)
(Elsevier, 2023-03-07)A 60-day feeding trial was conducted to evaluate the effect of inclusion frequency of hatchery-bred mud polychaete, Marphysa iloiloensis, as part of a combination diet on the reproductive performance and larval quality of domesticated P. indicus. The two treatments were fed with a combination of formulated diet, mussel, squid, and mud polychaete with once (T1) and twice (T2) a week inclusion in the feeding regime. In contrast, the control group was fed only a diet combination of mussels, squid, and formulated diet. There was no significant difference in the mean survival of male and female broodstock in all treatments upon termination (\(p\gt0.05)\). Shrimp in T1 and T2 performed significantly better than the control in terms of spawning frequency (\(p=0.037)\) and fertilization rate (\(p=0.001)\), while T1 had significantly high re-maturation (\(p=0.038)\). The survival, spawning, hatching rate, egg, and nauplii production did not differ significantly among treatments (\(p\gt0.05)\). Also, no difference in the gonado-somatic index (GSI), hepato-somatic index (HSI), spermatophore weight, and sperm count was found among diet treatments (\(p\gt0.05)\). Post larvae from T1 and T2 performed significantly better than the control after 25 (\(p=0.008)\) and 30 (\(p=0.009)\) minutes of abrupt exposure to freshwater, respectively. Therefore, even once weekly, mud polychaete inclusion is sufficient to improve the reproductive performance of P. indicus broodstock and produce offspring with boosted resistance in the hatchery. -
Evaluation of community-based livelihood development initiatives: The sustainable livelihood approach
(University of the Philippines Visayas, 2022)The sustainable livelihood approach (SLA) is a tool for evaluating livelihood strategies that utilize assets categorized as environmental, human, physical, financial and social assets. In this study, the SLA was applied to determine the main factors that influence the adoption of integrated multi-trophic aquaculture (IMTA) in milkfish (Chanos chanos Forsskål) farming among fisherfolks. The community-based field trials conducted in Nueva Valencia in Guimaras province for 4 years since 2015 revealed that stakeholders were receptive to IMTA and were able to situate its effects on livelihood assets. The assessment of 52 stakeholders showed small but significant improvements in most livelihood assets during the 3-phased evaluation period. Using the 5-scale Likert rating, the environmental livelihood assets were rated highest but showed slight significant decline due to perceived weather and water level changes that may have negatively affected fish production and income. Financial livelihood assets were initially rated low but significantly improved due to supplemental income derived from wages and harvest. The human livelihood assets significantly improved due to sustained hands-on aquaculture experience. Social livelihood assets also improved due to cohesive interaction during projects implementation. In contrast, the acquisition of physical livelihood assets did not significantly improve because these assets were not commensurate with the many stakeholders who would like to have access to these shared assets. Overall, stakeholders expect IMTA’s potential environmental and economic benefits to overcome additional adoption costs. These include the cost of physical and financial investments in seeds of co-culture species and modified enclosures. Stakeholders expect institutional and government support on further studies on the technical details of IMTA. Fisheries and aquaculture regulations should embed the necessary support systems for the adoption of IMTA and ensure a positive public perception of aquaculture. -
Innofloc: Innovative biofloc technology for the nursery production of shrimp, Litopenaeus vannamei in tanks
(University of the Philippines Visayas, 2022)Nursery systems are valuable production tools in shrimp aquaculture and are effective biosecurity facilities for the high-density culture of shrimp postlarvae (PL), resulting in healthy and uniform-sized juveniles. The nursery production of shrimp is usually carried out in small ponds; however, the use of small and circular tanks with plastic liners is gaining popularity. From an industry standpoint, there is a need to assess how nursery systems can improve the productivity of the shrimp production cycle. Hence, the use of small circular tanks coupled with the incorporation of biofloc technology was assessed in terms of its viability during the nursery production of the Pacific whiteleg shrimp, Litopenaeus vannamei. A 450m2 plastic-lined circular tank was installed and prepared for the stocking of L. vannamei postlarvae (PLs) at a density of 500 PLs per m2. Biofloc was produced and maintained throughout the nursery phase by adding brown sugar as a carbon source at carbon to nitrogen (C:N) ratio of 10. Daily monitoring of the various water quality parameters was carried out, while presumptive Vibrios were enumerated weekly. The shrimp juveniles were sampled for their body weight on the 14th day post-stocking and weekly until harvest on the 30th day of culture. The different water quality parameters were within optimum levels required for shrimp growth. Presumptive Vibrios were dominated by the yellow colonies. The shrimp attained 100% survival with an average body weight of 1.26 g and a feed conversion ratio (FCR) of 0.43 at the end of the nursery production phase. Our results indicate that the use of small circular tanks with biofloc during the nursery production phase of whiteleg shrimp is feasible and can be incorporated during the grow-out culture for improved and continuous production of this shrimp species. -
Bacterial microbiota of hatchery-reared freshwater prawn Macrobrachium rosenbergii (De Man, 1879) and their pathogenicity
(University of the Philippines Visayas, 2022)Quantitative and qualitative analyses of bacterial microbiota associated with hatchery of Macrobrachium rosenbergii (de Man, 1879) were conducted over three larval cycles, along with important water quality parameters. Physicochemical parameters (temperature, DO, salinity, pH, and total ammonia) of culture water were within the optimum level required for the hatchery phase of M. rosenbergii. Significant differences in aerobic plate count (APC) among larval stages were observed. Ten genera and 17 species were identified with the predominance of Gram-negative bacteria constituting 63 % of all isolates (n = 706). A total of 17 dominant bacterial isolates randomly selected from freshwater prawn larvae were screened for in vitro hemolysin activity using blood agar plate (BAP) medium with 5% sheep blood. Pathogenicity of the isolated bacterial strains was assessed through in vivo infection bioassay. Out of the 17 isolates tested, six isolates showed α – β zone of hemolysis on the BAP, i.e., Aeromonas caviae, Bacillus cereus, Plesiomonas shigelloides, Staphylococcus warneri, Vibrio cholerae non-01, and Vibrio furnissii, were used in the infection bioassay. The current data indicate that the six bacterial isolates tested, particularly B. cereus and V. cholerae non-01, are opportunistic pathogens capable of inducing disease epizootics, particularly in freshwater prawn hatcheries with suboptimal rearing water conditions. Thus, control measures to reduce the influx of pathogenic microbes in the system by maintaining good water quality and good farm management practices through disinfection of culture facilities, rearing water, and Artemia cysts, the practice of good hygiene of personnel, regular water exchange, and feed regulation, among others, and cautionary use of antibiotics, can be adopted. -
Characterization of wild eucheumatoids from Visayas, Philippines as inferred from the mitochondrial cox2-3 spacer sequence
(University of the Philippines Visayas, 2022)Eucheumatoids are the major seaweed species cultivated in most coastal areas in the Philippines, being a major source of income for many families. Seaweed farmers face issues such as lack of good quality cultivars and the presence of diseases and epiphytes in the cultivated stocks. This study characterizes the wild eucheumatoids, which are potential sources of cultivars that might be resistant to diseases and epiphytes. Wild eucheumatoids were collected from Iloilo (N=18), Guimaras (N=10), and Bohol (N=20), Philippines. Gross morphology was described, and a 10-mm apical tip was excised for DNA extraction. PCR amplification and sequencing using cox2-3 spacer genetic markers were done. Based on sequence data, 5 species can be identified from the Visayas region: the commercially cultivated Kappaphycus alvarezii (Iloilo, Bohol), K. striatus (Iloilo, Guimaras), and Eucheuma denticulatum (Iloilo, Bohol) and the currently uncultivated K. cottonii (Iloilo) and Kappaphycus sp. (Iloilo, Bohol). Haplotype network analysis identifies individuals of K alvarezii and E. denticulatum as haplotype E3 and E13, respectively, which are haplotypes previously reported from seaweed farms and globally distributed for cultivation purposes. The K. striatus from Iloilo and Guimaras are identified as haplotype E89, and the K. cottonii from Iloilo is haplotype E108 which has been described previously from seaweed farms and wild populations, respectively. Interestingly, Kappaphycus sp. from Iloilo and Bohol are identified as haplotype GUI1 initially described in 2014 and yet to be identified up to the species level. Our study shows that purely morphological identification is not sufficient to reliably identify species, thus highlighting the need to employ molecular techniques supporting strain selection. In summary, it reveals a rich diversity of wild eucheumatoids in the Visayas region of the Philippines. In-vitro studies need to reveal the performance of individuals from these wild populations but highlight a great potential for the development of new cultivars. -
Bacterial diseases of tilapia, their zoonotic potential and risk of antimicrobial resistance
(Wiley, 2023-03-05)Tilapia culture is an important source of income and nutrition to many rural families. Since 2000, the production of tilapia increased and reached domestic and global markets. Major farmed species is Nile tilapia (Oreochromis niloticus), in earthen ponds and cage cultures. Intensification contributed to global tilapia disease outbreaks, with bacterial infections causing mortalities and morbidities, threatening sustainable production. At tilapia farms, high nutrient concentrations, water temperature and fish densities enhance bacterial growth including virulent bacterial clones and potential zoonotic bacteria. Global warming favours this. This review respectively provides a comprehensive overview of the most common and emerging bacterial pathogens, diseases, clinical presentations and diagnostics of tilapia, including bacteria and diseases with zoonotic potential. First, common bacterial disease outbreaks, including streptococcosis, motile Aeromonas septicaemia, francisellosis, columnaris disease and vibriosis are described. Then, information on emerging bacterial infections of concern for tilapia, like edwardsiellosis through Edwardsiella ictaluri and E. tarda, as well as Aeromonas schubertii is provided. Reports of infectious bacterial tilapia disease outbreaks from other bacteria, including Lactococcus garvieae, Aerococcus viridans, Pseudomonas spp., Mycobacterium marinum and Chlamydia spp., and others are reviewed. Furthermore, bacteria with zoonotic potential, like Streptococcus agalactiae ST283, S. iniae, Aeromonas sp., E. tarda, Vibrio vulnificus pathovar (pv) piscis and M. marinum are included in the review, to provide the most current overview of the disease risks affecting production and post-harvest stages. Additionally, the status and risks of antimicrobial resistance in bacteria from tilapia and other cultured fish through imprudent use of antibiotics, and its future at a global level are provided. -
Improving artisanal fisheries through community-based prawn stock enhancement in a mangrove estuary
(Japan International Research Center for Agricultural Sciences, 2022)Tropical seafood, especially prawns, have always been in high demand with high market prices. Because of this, wild prawn resources are often overfished as indicated by the increasing number of fishers, but with overall declining catch volume and sizes through the years. Prawn aquaculture has, as of course, significantly contributed to the total yield in the past decades. However, the expansion of farms and ponds has often negatively affected the natural nursery grounds in the estuaries and mangroves. The economic gains from aquaculture are limited only to those who invested. However, local communities and artisanal fishers are often deprived of such gains. They are still reliant upon the dwindling natural stocks of fisheries resources. This is where the concept of stock enhancement comes in, wherein hatchery-produced juveniles are released into the water body in order to increase capture fisheries yield for local fishers. In this talk, I will present the case of the tiger prawn Penaeus monodon stock enhancement in the New Washington Estuary (NWE), in Aklan, central Philippines. The NWE was a productive fishing ground providing >24 kg/gear/day of catch in the 1970s but steadily declined to <0.7 kg/gear/day in 2010s. Prawn catches declined both in quality and quantity. Meanwhile, mangrove cover was reduced from 4800 ha in 1950s to only <800 ha in 2010s. Site-specific studies were conducted in NWE on prawn nursery rearing, small-scale release and monitoring experiments, as well as social assessments. Our studies produced essential scientific data to guide future stock enhancement activities in NWE. Even at an experimental scale, our releases showed significant increases in catch by at least 8% but can potentially increase daily income by >400%. However, in order to sustain the prawn fisheries and optimize the benefits, long-term management plans by all stakeholders are needed, especially between the communities and the local government. -
Seaweed Gracilariopsis heteroclada, formulated flaked diet, and a combination of both as feed for the abalone Haliotis asinina: Effect on growth and survival
(Springer, 2023-01-27)This paper compares the effect of feeding the abalone Haliotis asinina with seaweed (SW), formulated (FD), and mixed (SW+FD) diets. The feeding experiment, in six replicates, was conducted in a tank facility and lasted for 10 months. At the end of the experiment, abalone fed mixed diets had significantly the highest mean shell length and body weight (45.90±0.19 mm, 22.82±0.31 g), followed by those fed seaweeds (43.47±0.17 mm, 18.25±0.26 g), and lastly, by those fed formulated diets (41.78±0.18 mm, 15.76±0.22 g). Mean growth rates were significantly higher in abalone fed mixed diets at 1.48±0.03 mm and 1.69±0.04 g mo−1 than those fed seaweeds (1.26±0.03 mm, 1.27±0.06 g mo−1) or formulated diet (1.06±0.09 mm, 1.00±0.11 g mo−1). Survival was consistent 1 month from the start of culture until harvest—highest in abalone fed mixed diets (78.81±2.48%), followed by those fed seaweeds (70.12±4.07%), and lastly formulated diet (64.53±4.61%). Moreover, feed conversion ratios (FCR) were significantly lower in abalone fed mixed diets (15.48±0.69) and formulated diet (18.07±3.50) compared with those fed seaweeds (41.31±2.36). The body weight to shell length ratios (BW:SL), which were the same in all treatments at the start of the experiment (0.19 g mm−1), increased to 0.51, 0.44, and 0.38 in abalone fed mixed diets, seaweeds, and formulated diet, respectively. Based on the results of this experiment, giving abalone a mix of formulated and natural diets is the best feeding regime. The importance of a balanced and more nutritional diet on the well-being of the abalone was observed in this study.
